I connected my Ps4 Pro to a computer monitor without its own audio output. Now I wanted to look at a Bluray, but of course I could not change the volume. So I looked for a loudspeaker and found it. My question now: Could the speaker (has a USB cable with me) s.den USB ports, where to charge controller, connect and if so, would it work?

Yes, but plug it in the back as it also has a USB port. Then you can continue to charge your controller undisturbed.
